What Is A Sibling DNA Test
A Siblingship (or Sibship) DNA test analyzes the DNA of the tested individuals to determine whether the test participants have one or both of the parents in common, or if they are not related at all. Our Sibling DNA test includes testing for two siblings (bother/sister), but you may add more as needed; for instance a third sibling, or parents. Like a Paternity test, the mother is not included in the test, but can be added if the client desires. Adding a known parent to a Siblingship DNA test will greatly increase the conclusiveness of the test and is recommended if possible.
A Sibling DNA Test Can Answer 3 Questions:
- Do the Siblings share any common parent? (Are they related at all?)
- Do the Siblings share one common parent? (Half-Siblings)
- Do the Siblings share both parents? (Full Siblings)

Essentially, every human is composed of 46 paired chromosomes. In a scenario where two siblings are trying to determine if they share a common father, if the mother is tested along with the siblings, the known parent (mother in this case) would be compared to her child(ren) and thus the genes contributed by the mother would be identifed and separated. The remaining genes would be paternal, and can be much more easily compared to one another to determine if the siblings share the same father or not.
Understanding Siblingship or Sibship Testing:
In a Siblingship analysis, our DNA laboratory will analyze 16 of the 23 pairs of chromosomes for each of the siblings. Based on the type of genetic material inherited by each sibling, a siblingship index is determined. The higher the index is, the greater the likelihood that a true biological relationship is shared between the test participants. If the siblingship index is less than 1.00, this indicates that a biological relationship is less likely. If the siblingship index is greater than 1.00, this indicates that the two tested individuals are more likely to be biological siblings. To analyze the possibility that the siblings share only one common parent versus two common parents (mother or father), a half siblingship index is obtained. The ratio of the half siblingship index versus the full siblingship index would provide you with an idea of whether the two individuals are more likely to share only one common parent (e.g. mother only) or two common parents (mother and father).
Your Sibship test result will also provide a percentage of probability of the tested individuals being full siblings, half siblings and/or not related at all. These percentages are probably the easiest means to understanding your result. Each percentage will represent the probability that a true biological relationship does or does not exist.
How To Make Your Test More Conclusive:
With the exception of identical twins, each person’s DNA is as unique as their fingerprint. When the issue of siblingship is in question, the recommended method to determine whether individuals are true biological siblings is to test the parents along with the siblings. In situations where the alleged parents are unavailable for testing, a DNA siblingship test can still be performed to analyze the relationship of biological siblings.
A DNA Paternity or Maternity test will always provide a conclusive result, however Siblingship tests are different. Paternity and Maternity tests look for “obligatory” genes. “Obligatory” genes are genes that must be present to positively establish a relationship. When a Siblingship test is conducted, there are not obligatory genes. Therefore, including the mother(s) in a Siblingship test greatly increases the conclusiveness of the DNA analysis and is recommended if at all possible.
Additionally, if both alleged siblings are male, a Y-STR Test can be done to determine with great accuracy, whether the two males are from the same male lineage. Although the sibling DNA test may not always provide you with a conclusive answer, it may be able to provide you with an indication of whether you are more likely to be or not to be the true biological sibling of the other tested individual. If the reason for your siblingship DNA test is solely for curiosity, then our home non-legal sibling DNA test is right for you.
